The Institute for Biochemistry and Molecular Biology at RWTH Aachen University has an opening starting November 1st 2019 for a PhD Student (m / f / d) in Biochemistry / Molecular BiologyThe position is a part-

time position (65% h / w). The salary correspond to level 13 TV-L and is project-related offerd on a temporary contact for 2 years, with the option of extension.

The start date for the position is at November 1st 2019. The successful candidate will have the opportunity to pursue a doctoral degree.

Job description Project title : Function and Regulation of the mono-ADP-ribosyltransferase TARG1 in the Nucleolus. TARG1 is a 17 kDa protein, composed of a macrodomain fold and a short unstructured N-

terminal tail. Its macrodomain fold allows TARG1 to recognize and remove ADP-ribosylation, a post-translational protein modification.

ADP-ribosylation is catalyzed in cells by members of the ADP-ribosyltransferase diphtheria toxin-like (ARTD) family, which transfer either a single ADP-

ribose unit to a substrate (mono-ADP-ribosylation) or form ADP-ribose polymers on a substrate (poly-ADP-ribosylation) (Lüscher et al.

2018). Loss of TARG1 has been associated with ribosome biogenesis stress and neurodegeneration in early childhood. TARG1 interacts with ribosomal proteins and RNA processing factors in nucleoli (Bütepage et al.

2018). In agreement, we found that TARG1 has RNA binding activity. Based on a computational model of a TARG1-RNA complex, we identified the relevant interaction surface in TARG1.

The aim of this proposal is to investigate the single or combined contributions of TARG1’s RNA binding and ADP-ribosylhydrolase activity to its nucleolar function.

    Required are very good communication and presentation skills, both in German and in English. We expect excellent experimental skills and experience in all standard procedures involving the analysis of proteins, ribonucleic acids and cells.

    We offer You will be part of a highly interactive, international team. You will have access to all relevant equipment and a broad range of core facilities, including proteomics and next generation sequencing.

    You will participate in progress reports, journal clubs and seminars on a weekly base. You will present your work at international meetings.
